SS-31 peptide, also known as Elamipretide, is a synthetic tetrapeptide (amino acid sequence: D-Arg-Dmt-L-Lys-D-Ala) designed

to target and protect mitochondrial membranes. First identified for its ability to interact with cardiolipin (CL), a phospholipid

exclusively located in the inner mitochondrial membrane, SS-31 has been studied for its role in mitigating mitochondrial

dysfunction associated with various diseases.

The peptide's structure features a cyclic conformation that allows high-affinity binding to cardiolipin, a key lipid critical for

maintaining mitochondrial membrane integrity and function. By binding to cardiolipin, SS-31 stabilizes the mitochondrial inner

membrane, reduces oxidative stress, and inhibits mitochondrial permeability transition pore (mPTP) opening-an event linked to

mitochondrial dysfunction and cell apoptosis. Additionally, it protects mitochondrial electron transport chain complexes, enhancing
